Research Focus (Non-Clinical)
5-amino-1MQ is a membrane-permeable small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), a cytosolic enzyme that methylates nicotinamide using S-adenosylmethionine (SAM). In cultured adipocytes, methylquinolinium NNMT inhibitors were associated with reduced intracellular 1-methylnicotinamide (1-MNA), increased intracellular NAD+ and SAM, and suppressed lipogenesis. In the same study, selectivity screening found no inhibition of structurally related SAM-dependent methyltransferases or of NAD+ salvage-pathway enzymes. It is presented here as a reference compound in NNMT enzymology and NAD+/SAM metabolic-flux research. Findings described here are from cell-culture and mouse model systems.
